摘要: A process for the effective extraction and fractionation of phlorotannins from Fucus vesiculosus with high antioxidant potentials was investigated. The activity F. extract/fractions assessed by 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging, reducing power, ferrous ion-chelating assays. Among crude extract different polarity fractions, phlorotannin-enriched ethyl acetate fraction possessed highest DPPH scavenging power. This further fractionated Sephadex LH-20 column chromatography or ultrafiltration. properties were evaluated both above chemical tests a mononuclear cell-based bioassay. subfractions LH-2 LH-3 total phlorotannin content exhibited strong quenching activity, comparable to those ascorbic acid butylated hydroxytoluene significantly higher than that α-tocopherol. Polyphenols in found consist mainly molecular weight polymers. There no clear relationships between degree polymerization, size, activity. All separated ultrafiltration showed ability scavenge reactive oxygen species generated cells. Further characterization compounds performed on six subfractions. Several oligomers tentatively identified basis HPLC-ESI-MS(n) analyses.
