摘要: Interactions between polysaccharides and phospholipid bilayers have already been demonstrated in the literature but little is known about influence of macromolecule conformations related to solvent characteristics (pH, ions, ionic strength). In this study we investigated conformation iono- thermo-sensitive polysaccharides, iota- kappa-carrageenans, their interaction with a dimyristoylphosphatidylcholine (DMPC) model bilayer. The was performed two different media (NaCl 150 mmol/L, pH 6.5, NaCl 300 mmol/L, 6.5). first part, kappa-carrageenan samples characterized by size exclusion chromatography (SEC) coupled multi-angle laser light-scattering detector (MALLS). SEC–MALLS results clearly show polysaccharide chain association at high strength. second polysaccharide–membrane has studied, using fluorescent probes embedded membrane. thermotropic properties membrane were fluorescence depolarization 1-(4-trimethylammonium-phenyl)-6-phenyl-1,3,5-hexatriene (TMA-DPH). surface accessibility evaluated quenching 2-(9-anthroyloxy) stearic acid (2-AS). Whatever strength tested, presence notably enhances fluidity below Tm. This sign an polar level more marked low concentration. contrast, liposomes bilayer drastically lowered when increasing induced macromolecular adsorption on liposome surface, enhanced association. An enhancement induces conformational modification chains which modifies ability interact Copyright © 2001 John Wiley & Sons, Ltd.
