摘要: Glycerol is widely used to stabilize cytochrome P-450 and prevent its transformation P-420. The effect of glycerol on the content activity human P-4502E1 (CYP2E1) in a HepG2 cell line that stably constitutively expresses this was evaluated by immunoassays oxidation p-nitrophenol. Addition 100 200 mM culture medium resulted 2 1/2- 3-fold increase CYP2E1 microsomes isolated from cells. Increases could be observed within 4 8 hr after addition medium. had no b5 or activities NADPH-cytochrome reductase NADH-cytochrome reductase. Upon cycloheximide stop protein synthesis, decreased with apparent half-lives 6 hr, respectively. prevented loss activity. Labeling [35S]methionine, followed pulse-chase experiments cold methionine immunoprecipitation indicated half-life for approximately 3 hr. increased 11 Stabilization not additive synergistic ethanol 4-methylpyrazole, suggesting all three agents elevate similar type mechanism model. These results indicate can interact it against proteolytic degradation, increasing enzyme thereby elevating CYP2E1.
