摘要: Immunohistochemistry has rapidly acquired an important position in present-day pathology laboratories. The expanding selection of monoclonal and poly-clonal antibodies with well defined specificities greatly increased the potentially obtainable information from routine biopsy specimens. Basically, only limiting factors for demonstration presence any biologically significant, antigenic molecules are specificity antibody on one hand and, other, tolerance epitope(s) to various methods required handling Appropriate characterization used, whether polyclonal or is imperative. They should be thoroughly analysed lack cross reactions electrophoretic blots react corresponding antigen. On target side, overall best results obtained frozen sections, fixed instance acetone. Alcohol Bouin fixation followed by paraffin embedding also gives generally good results, although long-term preservation material less good. Formalin embedded can successfully applied many instances sensitivity detecting certain antigens including intermediate filaments markedly decreased. Enzyme pre-treatment such sections often improve probably degrading cross-links that may mask sites molecules. Inclusion proper controls into each diagnostic run course mandatory. technical aspects have been considered several excellent reviews (e.g. Taylor 1978; Lewis et al. 1983).
