Aberrant Monoaminergic System in Thyroid Hormone Receptor-β Deficient Mice as a Model of Attention-Deficit/Hyperactivity Disorder

摘要: Background Thyroid hormone receptors are divided into 2 functional types: TRα and TRβ. play pivotal roles in the developing brain, disruption of thyroid can produce permanent behavioral abnormality animal models humans. Methods Here we examined behavioralchanges, regional monoamine metabolism, expression epigenetic modulatory proteins, including acetylated histone H3 deacetylase, brain TRα-disrupted (TRα (0/0) ) TRβ-deficient (TRβ (-/-) mice. Tissue concentrations dopamine, serotonin (5-hydroxytryptamine) their metabolites mesocorticolimbic pathway were measured. Results TRβ mice, a model attention-deficit/hyperactivity disorder, showed significantly high exploratory activity reduced habituation, whereas mice normal activity. The biochemical profiles dopamine 5-hydroxytryptamine low metabolic rates caudate putamen nucleus accumbens overall but not Furthermore, was dorsal raphe deacetylase 2/3 proteins widely increased mesolimbic system. Conclusions These findings suggest that deficiency causes dysfunction monoaminergic system, accompanied by during maturation process.
