Wnt signaling induces differentiation of progenitor cells in organotypic keratinocyte cultures

摘要: Interfollicular skin develops normally only when the activity of progenitor cells in basal layer is counterbalanced by exit into suprabasal layers, where they differentiate and cornify to establish barrier function. Distinct stem compartments have been demonstrated hair follicles sebaceous glands, but there are few data describe control interfollicular cell activity. Wnt signaling has shown be an important growth-inducer many other tissues. Here, we test effect ectopic Wnt1 expression on behavior organotypic culture model, find that inhibits their growth promotes terminal differentiation. These results consistent with phenotypes reported for transgenic mice engineered gain or loss function skin, which would recommend our model as accurate one molecular analysis. Since it known canonical ligands expressed likely this pathway regulates balance differentiation, suggests could pathogenesis.
