Circulating miRNAs associated with bone mineral density in healthy adult baboons.

摘要: MicroRNAs (miRNAs) regulate gene expression post-transcriptionally and circulate in the blood, making them attractive biomarkers of disease state for tissues like bone that are challenging to interrogate directly. Here we report on five miRNAs - miR-197-3p, miR-320a, miR-320b, miR-331-5p, miR-423-5p associated with mineral density (BMD) 147 healthy adult baboons. These baboons range age from 15 25 years (45 75 human equivalent years) were 65% female a broad BMDs including minority osteopenic individuals. generated via RNA sequencing buffy coats collected at necropsy areal BMD evaluated DXA lumbar vertebrae post-mortem. Differential analysis controlled underlying pedigree structure these animals account genetic variation which may be driving miRNA abundance values. While many have been risk osteoporosis, this finding is interest because cohort represent model normal aging metabolism rather than cohort. The replication associations osteoporosis or other metabolic disorders suggests an overlap states. We suggest involved regulation cellular proliferation, apoptosis, protein composition extracellular matrix throughout life. However, age-related dysregulation systems lead causing among individuals clinically defined disease.
