摘要: Stimulation of human polymorphonuclear leucocytes (PMN) with phagocytosable particles [yeast-IgG (Y-IgG)], lipopolysaccharide (LPS), tumour necrosis factor (TNF) or formyl-methionyl-leucyl-phenyl-alanine (FMLP) results in an increase the interleukin-8 (IL-8) mRNA accumulation and a subsequent release protein. Here, we report that interferon-gamma (IFN-gamma) down-regulates constitutive IL-8 levels expressed by resting PMN. As shown Northern analysis, this down-modulation occurred rapidly, was not dependent on new protein synthesis, caused increased rate degradation mRNA. Preincubation PMN IFN-gamma significantly inhibited their ability to upon stimulation TNF, LPS, FMLP Y-IgG, but enhanced respiratory burst capability response TNF. TNF-, LPS- FMLP-induced expression also selectively IFN-gamma. Taken together these findings suggest has important regulatory effects acute inflammatory because its capacity modulate negatively gene secretion Further observations revealed that, PMN, is finely regulated, cycloheximide (CHX), inhibitor super-induces for dose- time-dependent manner.
