摘要: Abstract During acute inflammation, the first line of cellular response for host defense is neutrophil. In addition to historic role neutrophil as a phagocyte, recent studies have identified this cell an important source number cytokines. study, we provide evidence that significant interleukin-8 (IL-8). Neutrophils freshly isolated from whole blood were not found constitutively express IL-8 mRNA. contrast, when these leukocytes cultured on plastic they activated, leading expression de novo steady-state levels addition, neutrophils treated with cycloheximide, there was "superinduction" mRNA and inhibition antigenic production. subsequently stimulated lipopolysaccharide (LPS), tumor necrosis factor-alpha, or interleukin-1-beta antigen in both time- dose-dependent manner. Furthermore, traditional chemotactic/activating factors, such split product fifth component complement (C5a), formylmethionyleucylphenylalanine (fMLP), leukotriene B4 (LTB4) manner did produce IL-8, compared unstimulated controls. exposed either agonists presence LPS, production significantly elevated, stimuli alone, suggesting synergistic response. These data would suggest can no longer be viewed only phagocyte warehouse proteolytic enzymes, but pivotal effector able respond mediators its environment generate This latter may elicitation additional orchestrate conventional immune at sites inflammation.
