Micropropagation of the critically endangered Western Australian species, Symonanthus bancroftii (F. Muell.) L. Haegi (Solanaceae)

摘要: A micropropagation protocol was developed for the conservation of critically endangered Western Australian shrub,Symonanthus bancroftii. It necessary to screen antioxidant treatments prevent occurrence lethal browning explants upon excision. Potassium citrate and citric acid (0.1% w/v in a 4:1 ratio) prevented oxidative superior untreated control or other tested. Half strength Murashige Skoog (MS) medium containing 0.5 μM kinetin 0.25 benzyladenine produced three-fold multiplication compared 1.75×, 1.5×, 1.8× 1× 2.5 + benzyladenine, 5 gibberellic acid, 1 3 half MS with no plant growth regulators, over 4 weeks. Root production achieved indole-3-butyric (IBA) α-naphthaleneacetic (NAA) at 0.5/0.5 (31% rooting) 1.0/1.0 (36% rooting), after four Paclobutrazol (PBZ) 0, 3.4 (1 mg 1−1), 10.2 (3 17 (5 1−1) improved tolerance desiccation transfer ofin vitro rooted shoots soil. PBZ increased survival 90% 50% those plantlets not treated PBZ. The acclimatisation period from glasshouse shadehouse week weeks without any number roots per shoot controls.
