Comparison of metoprolol and carvedilol pharmacology and cardioprotection in rabbit ischemia and reperfusion model.

摘要: Carvedilol, a selective α1 and non-selective β-adrenoceptor antagonist antioxidant, has been shown to provide significant cardiac protection in animal models of myocardial ischemia. To further explore the mechanisms contributing carvedilol cardioprotection efficacy, effects on hemodynamic variables, infarct size myeloperoxidase activity (an index neutrophil accumulation) were compared with β1-selective adrenoceptor antagonist, metoprolol. Carvedilol (1 mg/kg) or metoprolol mg/kg 1 mg/kg+0.5 90 min later) was given intravenously 5 before reperfusion. In vehicle-treated rabbits, ischemia (60 min) reperfusion (180 resulted increments left ventricular end diastolic pressure, large infarcts (59±2.6% area-at-risk) marked increase (0.59±0.09 U/100 mg tissue). treatment sustained reduction pressure-rate-index significantly smaller (22.0±2.5%, P<0.01 vs. vehicle) as well decreased (0.186±0.056 tissue, vehicle). The highest dose metoprolol, mg/kg, that comparable 1.0 carvedilol, failed reduce ischemic (35±3.1%) larger than carvedilol-treated animals. Taken together, this study suggests superior over is not consequence variances but possibly result additional pharmacological properties such antioxidant anti-neutrophil effects.
