摘要: Background: in vitro cytotoxicity testing provides a crucial means of ranking compounds for consideration drug discovery. The choice using particular viability or assay technology may be influenced by specific research goals. Objective: Although the high-throughput screening (HTS) utility is typically dependent upon sensitivity and scalability, it also impacted signal robustness resiliency to interferences. Further should given data quality, ease-of-use, reagent stability, matters cost-effectiveness. Methods: Here we focus on three main classes assays that are at present most popular, useful, practical HTS discovery efforts. These methods measure: i) metabolism reductase activities; ii) bioluminescent ATP assays; iii) enzymes ‘released’ into culture medium. Multi-parametric technologies briefly discussed.
