摘要: Spherical nucleic acid (SNA) constructs are promising new single entity gene regulation materials capable of both cellular transfection and knockdown, but thus far promiscuous structures, exhibiting excellent genetic little selectivity. In this communication, we describe a strategy to impart targeting capabilities these through noncovalent functionalization with complementary antibody-DNA conjugate. As proof-of-concept, designed HER2-targeting SNAs demonstrated that such structures exhibit cell type selectivity in terms their uptake, significantly greater knockdown cells overexpressing the target antigen as compared analogous antibody-free off-target materials.
