Screening and identification of a gene, PSE-1, that affects protein secretion in Saccharomyces cerevisiae

摘要: Utilizing a screening method designed for the identification of genes involved with enhanced protein secretion in Saccharomyces cerevisiae we identified gene, which named PSE-1 (Protein Secretion Enhancer). Overexpression multi-copy plasmid, as shown by Northern hybridization, gave fourfold enhancement total secretion. The repertoire proteins that are found to be secreted greater quantities include three known biologically active proteins: k1 killer toxin, alpha-factor, and acid phosphatase. gene is located on chromosome XII yeast genome codes hydrophobic containing 1089 amino acids. Haploid cells contained LEU2 insertion mutation grow very poorly, phenotype similar other conditional SEC mutants at restrictive temperature.
