摘要: The technique of small-molecule microarray (SMM) screening is based on the ability small molecules to bind various soluble proteins. This type interaction easily detected by presence a fluorescence signal produced labeled antibodies that specifically recognize unique sequence (tag) present target protein. fluorescent intensity values are determined signal-to-noise ratios (SNRs). SMM high-throughput, unbiased method can rapidly identify novel direct ligands for protein targets. binding-based assay format generally applicable most proteins, but it especially useful targets do not possess an enzymatic activity. SMMs enable in purified form or context cellular lysate, likely providing more physiologically relevant environment.
