Effect of signal transduction pathways on the action of thapsigargin on rat mast cells: Crosstalks between cellular signalling and cytosolic pH

摘要: Abstract Thapsigargin elicits histamine release on rat mast cells, and this effect is increased if cells are pretreated with thapsigargin before the addition of external calcium. Okadaic acid does not modify response to thapsigargin, while sodium fluoride or phorbol esther 12- O -tetra-decanoylphorbol-13-acetate (TPA) increases several fold sensitivity thapsigargin. On other hand, pertussis cholera toxins inhibit activity Na + −H exchanger, being blocked by modified TPA. The metals cadmium lanthanum completely block TPA -H exchanger. influx 45 Ca in but treated calcium, markedly first 2 min returning normal. Our results indicate that exocytosis modulated crosstalks between intracellular cytosolic pH
