摘要: Rat liver regeneration was studied from 24 hours to 8 days after a single intraperitoneal injection of D-galactosamine (GalN). Morphological changes in the were analyzed parallel with sequential expression histone-3 mRNA (a marker cell proliferation), fetal alpha-fetoprotein (AFP) and gamma-glutamyl transpeptidase (GGT) (markers hepatocytes), albumin glucose-6-phosphatase (G6Pase) adult hepatocytes). Proliferation nonparenchymal epithelial cells (NPC), detected situ by [3H]thymidine labeling or expression, began primarily portal area around bile ducts. After 2 days, labelling intensity increased rows clusters NPC which expanded zone invaded into parenchyma. On 3 5, expressing his-3 further, forming pseudo-ducts islet-like structures (NPC structures). Proliferating positive for GGT. Some GGT also form AFP mRNA, lagged behind peaked on day 5. 3, some appearance expressed mRNA. Double label hybridization mRNAs dual histochemistry G6Pase showed simultaneous these markers Other 5 had morphological between hepatocytes (transitional cells). 2, reached maximum then declined. did not express These findings indicate that GalN injury, responds activation progenitor proliferate differentiate mature hepatocytes. Adult can GAlN but do undergo dedifferentiation/redifferentiation during hepatic lobule.
