摘要: Simvastatin, a cholesterol lowering drug, has been shown to have positive effects on fracture healing and bone regeneration based its dual effect; anabolic anti-resorptive. In this study the focus anti-resorptive effect of drug impact degradation acidic calcium phosphate cement. The was added pre-mixed cement in three different doses (0.1, 0.25 0.5 mg/g cement) release measured. Furthermore loaded cements osteoclast differentiation resorption evaluated by TRAP activity, number multinucleated cells, gene expression ion concentration vitro using murine marrow macrophages. simvastatin did not affect cell proliferation while it clearly inhibited osteoclastic at all as staining, activity expression. Consistent with these results, also impaired osteoclasts indicated reduced concentrations. conclusion, our findings suggest that simvastatin-doped inhibits mediated thus slowing down rate. addition simvastatin's makes cement-drug combination promising graft material, especially useful for sites compromised formation.
