摘要: Vascular adhesion protein-1 (VAP-1) is an molecule, which upon inflammation rapidly translocated from intracellular sources to the endothelial cell surface. We have recently discovered that sialic acid- binding immunoglobulin-like lectin 9 (Siglec-9) a leukocyte ligand of VAP-1 and 68Ga-labeled Siglec-9 motif peptide facilitates in vivo imaging inflammation. This study evaluated feasibility 68Ga-DOTA-Siglec-9 positron emission tomography (PET) for assessment synovitis. Rabbits with synovial were injected 18F-FDG or studied by gamma counting autoradiography. Certain rabbits also examined magnetic resonance (MRI). After PET imaging, intravenously administered anti-VAP-1 antibody evaluate luminal expression immunohistochemistry. Finally, positive vessels double staining rheumatoid arthritis synovium. Intra-articular injection hemagglutinin induced mild rabbit knee VAP-1. Synovitis was clearly visualized addition 18F-FDG-PET MRI.
