摘要: This study has used light and electron microscope immunohistochemical biochemical methods to localize characterize vitronectin in early bone formation of developing rat mandible with rabbit antimurine IgG. Developing jaws foetuses were collected at embryonic day 15 (day 15) 18 from pregnant Wistar rats. After aldehyde fixation, specimens without osmium post-fixation dehydrated embedded paraffin, Spurr's resin or LR gold for morphological examinations. At the level, samples, positive immunostaining was observed small elongated areas intercellular matrix osteoblasts. Concomitant initiation mineralization 16, staining similarly containing osteoblasts but not clearly detected fully mineralized matrix. The same profile days 17 18. ultrastructural immunogold particles over unmineralized cisterns rough-surfaced endoplasmic reticulum Golgi apparatus as well demineralized 16-18. In order assess presence mineral phase, mineral-binding proteins extracted fresh using a three-step technique: 4M guanidine HCl (G1 extract), aqueous EDTA (E followed by HCl. Subsequent Western blot analysis sodium dodecyl sulphate (SDS)-polyacrylamide gel electrophoresis revealed that antibodies produced only single band an M(r) approximately 73000 both G1 E extracts, indicating These results indicate that, onset formation, synthesize release vitronectin, which is subsequently incorporated into becomes specific component tissues. observation these critical stages suggests it may be involved regulation formation.
