Effects of canthin-6-one alkaloids from Zanthoxylum chiloperone on Trypanosoma cruzi-infected mice

摘要: Canthin-6-one (1), isolated from Zanthoxylum chiloperone (Rutaceae), possesses a broad sprectum of antifungal and leishmanicidal activities. In this study, we have examined the antiparasitic effects canthin-6-one 5-methoxycanthin-6-one (2), N-oxide (3), as well that total alkaloids stem bark, in Balb/c mice infected either acutely or chronically with Trypanosoma cruzi. The compounds were administered orally subcutaneously at 5 mg/kg/day for 2 weeks, whereas alkaloidal extract was given 50 weeks. activity compared benznidazole case acute infection, parasiteamia significantly reduced following oral treatment (1). Moreover, bark led to high levels parasitological clearance. Seventy days post-infection, serological response model different between (1) benznidazole-treated mice. Chronic disease showed both above dosage induced 80-100% animal survival untreated controls. These results indicate exhibits trypanocidal vivo mouse chronic infection. This is first demonstration anti-Trypanosoma cruzi member chemical group (canthinones). Considering very low toxicity our suggest long-term natural product could prove advantageous current chemotherapy Chagas disease.
