Enhanced transcriptome maps from multiple mouse tissues reveal evolutionary constraint in gene expression

摘要: Mice have been a long-standing model for human biology and disease. Here we characterize, by RNA sequencing, the transcriptional profiles of large heterogeneous collection mouse tissues, augmenting transcriptome with thousands novel transcript candidates. Comparison in cell lines reveals substantial conservation programmes, uncovers distinct class genes levels expression that constrained early vertebrate evolution. This core set captures fraction output mammalian cells, participates basic functional structural housekeeping processes common to all types. Perturbation these is associated significant phenotypes including embryonic lethality cancer. Evolutionary constraint gene not reflected genomic sequences, but conserved epigenetic marking, as well characteristic post-transcriptional regulatory programme, which sub-cellular localization alternative splicing play comparatively roles.
