Rapid Characterization of Chemical Components in Edible Mushroom Sparassis crispa by UPLC-Orbitrap MS Analysis and Potential Inhibitory Effects on Allergic Rhinitis.

摘要: Sparassis crispa is a kind of edible fungus widely grows in the north temperate zone, which shows various medicinal properties. Due to complexity chemical constitutes this species, few investigations have acquired comprehensive configuration for profile it. In study, strategy based on ultra-high performance liquid chromatography (UPLC) combined with Orbitrap mass spectrometer (MS) was established rapidly characterizing components S. crispa. Through summarized MS/MS fragmentation patterns reference compounds and systematic identification strategy, total 110 attributed six categories were identified first time. Moreover, allergic rhinitis (AR) worldwide inflammatory disease seriously affecting human health, development drugs treat AR has been topic interest. It reported that extracts showed obvious inhibitory effects degranulation mast cell- allergen-induced IgE proinflammatory mediators, but active specific mechanism still not clear. Src family kinases (SFKs) participate initial stage allergy occurrence, are considered targets treatment. Herein, basis self-built database, virtual screening applied predict potential SFKs inhibitors crispa, using known crystal structures Hck, Lyn, Fyn, Syk as receptors, followed by anti-inflammatory activity evaluation screened hits intracellular calcium mobilization assay. As results, sparoside A directly confirmed strong an IC50 value 5.06 ± 0.60 μM. This study provides useful elucidation composition demonstrated its AR, could promote research effective agents from natural resources.
