摘要: Alterations in polyamine metabolism may be a critical mechanism of monocrotaline (MCT)-induced structural remodeling the pulmonary vasculature. In present study, hypothesis that MCT, through induction oxidative stress, modulates cellular regulatory mechanisms which turn might involved upregulation fibronectin production artery endothelial cells (PAEC) was examined. A 24-h treatment with MCT significantly increased PAEC concentrations as compared to vehicle-treated cells. addition, exposure caused an increase abundance ornithine decarboxylase (ODC) mRNA, ODC activity and enhancement spermidine import into PAEC. Inhibition ofde novopolyamine synthesis further uptake MCT-treated The depletion contents blockade bothde biosynthesis transport prevented MCT-induced increases medium level fibronectin. stimulated stress determined by levels thiobarbituric acid reactive substances, enhanced dichlorofluorescein fluorescence activation NF-KB. co-treatment dimethylthiourea, oxygen radical scavenger, oxidation attenuated disturbances metabolism. These data suggest can stimulate processes possibly stress. study have significant implication understanding hypertension
