摘要: Myeloperoxidase (MPO), an abundant heme-containing enzyme present in neutrophils, monocytes, and macrophages, is produced high levels during inflammation, associated with poor reproductive outcomes. MPO known to generate hypochlorous acid (HOCl), a damaging reactive oxygen species (ROS) utilizing hydrogen peroxide (H2O2) chloride (Cl-). Here we investigate the effect of activated immune cells on oocyte quality. Mouse metaphase II oocytes were divided into following groups: 1) Incubation catalytic amount (40 nM) for different incubation periods presence 100 mM Cl- without H2O2 melatonin (100 μM), at 37°C (n = 648/648 total number each group cumulus cells); 2) Co-cultured mouse peritoneal macrophage neutrophils (1.0 x 106 cells/ml) absence (200 inhibitor/ROS scavenger, HTF media, 200/200); 3) Untreated incubated 4 hrs as controls 73/64). Oocytes then fixed, stained scored based microtubule morphology chromosomal alignment. All treatments found negatively affect quality time dependent fashion compared controls. In all cases offered no protection; however significant protection was by melatonin. Similar results obtained treated neutrophils.
