Microarray analysis and RNA silencing link fra-1 to cd44 and c-met expression in mesothelioma.

摘要: Malignant mesothelioma is a cancer with poor prognosis associated exposures to asbestos. The mechanisms of asbestos-induced mesotheliomas are unclear, and studies required find diagnostic tools therapies improve the survival rates patients. After oligonucleotide microarray analysis (Affymetrix array) normal rat pleural mesothelial (RPM) cells, RPM cells exposed crocidolite asbestos, mesotheliomas, subsets genes that changed in expression were categorized, including highly up-regulated, early response proto-oncogene, fra-1. Increases fra-1 both human subset common asbestos-exposed mimicked patterns subsequently confirmed using real-time quantitative PCR. Using RNA interference technology, gene silenced assayed by PCR for possible fra-1-regulated genes. Results reveal induction cd44 c-met causally linked expression, connecting governing cell motility invasion mesothelioma. These suggest inhibition signaling pathways may be strategy therapy malignant
