Glucocorticoid receptor isoforms and effects of glucocorticoids in ovulated mouse oocytes and preimplantation embryos

摘要: To investigate possible involvement of glucocorticoid receptor (GR) in mediating effects maternal stress or therapeutically administered glucocorticoids on early embryo, we analyzed the expression GR subtypes ovulated mouse oocytes and preimplantation embryos. RT-PCR analysis results showed that GRα GRγ transcripts are relatively highly expressed oocytes, both present at lower amounts We also detected low two other splice variants, GRβ a transcript orthologous to human GR-P subtype, mainly blastocyst stage. Using western blot analysis, several protein bands differed size between compare corticosterone (a major endogenous rodents) dexamethasone synthetic glucocorticoid) embryos, cultured embryos presence these glucocorticoids. Corticosterone strong inhibitory effect embryo development (starting from 50 μM concentration), without significant influence apoptosis incidence. On hand, induced cells 1.5 its was less detrimental than found with same dose corticosterone. In summary, our different composition changes during development. Moreover, differences development, which might be associated activation subtypes.
