Chimiokines et récepteurs de chimiokines : futures cibles dans le traitement des maladies allergiques

摘要: Resume Les chimiokines et leurs recepteurs constituent un reseau complexe de molecules qui jouent role essentiel dans les processus migration d’activation des leucocytes. Ce intervient le developpement la reaction inflammatoire allergique, en particulier par ses capacites a reguler trafic eosinophiles, basophiles, mais aussi lymphocytes Th2. De nombreux travaux experimentaux realises in vitro, ou vivo grâce aux modeles animaux, etudes portant sur biopsies bronchiques chez l’asthmatique confirment place preponderante devolue CCR3, CCR4 CCR8 (eotaxine, RANTES pour l’eosinophile, TARC, MDC l’I-309 Th2). Plusieurs voies therapeutiques interessantes apparaissent : anticorps anti-recepteurs, chemokines « modifiees » antagonistes synthetiques, tous capables degres divers bloquer l’inflammation allergique. Cependant si certaines substances apparaissent efficaces experimentation animale, aucun n’a encore atteint niveau l’utilisation clinique humaine.
