摘要: Techniques for creation of colon carcinoma epithelial cells lines in long-term culture have been available years, but these techniques involved mechanical or enzymatic methods to separate from surrounding tissues. While this practice has intermittently successful, the effect traumatic on cellular behavior is unknown. Samples patient volunteers were subjected serial nonenzymatic disruptions fibrous Cells collected, allowed proliferate, and then tested their characteristics (mucin, vimentin, cytokeratin, colon-specific antigen, carcinoembryonic antigen) by immunohistochemistry flow cytometry. Growth determined phase-contrast microscopy, multiple passage, freeze/thaw effects. Tumorigenicity was proven nude mice. Of 11 initial attempts, three resulted stable which are demonstrated behave similarly original tumors they derived. This technique adds another reliable vitro tool study carcinoma.
