摘要: Cytokines released from tumour cells may have function as signals to neighbouring immune and inflammatory cells. Several studies shown that the immunoregulatory cytokines IL-10 transforming growth factor-beta1 (TGF-beta1) well prostaglandin-E2 (PGE2) play an important role in tumour-induced immunosuppression. The aim of study was investigate effect colon carcinoma cell lines on production peripheral monocytes (PBMC) lamina propria mononuclear (LPMC). We examined four (HT-29, Caco-2, Colo-320 HCT-116) determined their TGF-beta1, PGE2. Peripheral were isolated by density gradient centrifugation LPMC surgical specimens using a collagenase digestion method. Monocytes cultured with conditioned medium or co-culture Supernatants then for ELISA assays. All stimulated produce markedly increased levels IL-10. Colon cancer secreted negligible IL-10, but high amounts TGF-beta1 Neutralization administration anti-TGF-beta neutralization PGE2 anti-PGE2 antisera reduced markedly, indicating cell-derived are major factors stimulation. In vitro stimulation could confirm at picogram concentrations able prime enhanced production. Our results demonstrate enhance ability intestinal macrophages monocyte act tumour-protecting mechanism impairing activation anti-tumour cytokines.
