Opioid receptors: Structural and mechanistic insights into pharmacology and signaling

摘要: Opioid receptors are important drug targets for pain management, addiction, and mood disorders. Although substantial research on these subtypes of G protein-coupled has been conducted over the past two decades to discover ligands with higher specificity diminished side effects, currently used opioid therapeutics remain suboptimal. Luckily, recent advances in structural biology provide unprecedented insights into receptor pharmacology signaling. We review here a few studies that have crystal structures as basis revealing mechanistic details signal transduction mediated by receptors, purpose discovery.
