摘要: An improved method for the rapid and sensitive determination of endogenous dopamine (DA) 3,4-dihydroxyphenylacetic acid (DOPAC) in superfusates single rat striatal slices, using high-performance liquid chromatography (HPLC) coupled with 'coulometric' electrochemical detection (EC), is described. Superfusates are directly injected into an HPLC-EC system following addition a small aliquot solubilized ascorbate-oxidase. DA DOPAC both separated quantitated 3-5 min. Twelve to 15 samples can be analyzed each hour nominal limit 1.0 pg per injection compound or 10-20 pg/ml superfusate. The present was used study changes outflow overflow slices electrical field stimulation, absence presence catecholamine uptake inhibitor nomifensine. Studies 1 min superfusate collections clearly showed that stimulation produced latent increase as compared DA. routine sensitivity sample throughout allows studies DOPAC, well involving time-dependent these compounds.
