摘要: Genome rearrangements can take place by a process known as replication slippage or copy-choice recombination. The occurs between repeated sequences in both prokaryotes and eukaryotes, is invoked to explain microsatellite instability, which related several human diseases. We analysed the molecular mechanism of short direct repeats, using vitro single-stranded DNA template that mimics lagging strand synthesis. show involves polymerase pausing, must within repeat, pausing dissociates from DNA. also present evidence that, upon dissociation, only terminal portion newly synthesized separates anneals another repeat.
