摘要: Skeletal erosion has been found to represent an independent prognostic indicator in patients with advanced stages of chronic lymphocytic leukaemia (CLL). Whether this phenomenon also occurs early CLL phases and its underlying mechanisms have yet be fully elucidated. In study, we prospectively enrolled 36 consecutive treatment-naive analyse skeletal structure bone marrow distribution using a computational approach PET/CT images. This evaluation was combined the analysis RANK/RANKL loop activation leukemic clone, given recent reports on role progression. Bone particularly evident long shafts, progressively increased from Binet stage A C, correlated both local expansion metabolically active documented by FDG uptake number RANKL + cells present circulating blood. immune-deficient NOD/Shi-scid, γcnull (NSG) mice, administration cells caused appreciable compact that prevented Denosumab. cell proliferation vitro RANK expression impaired Denosumab-mediated disruption loop. study suggests interaction between stromal elements able simultaneously impair increase proliferating potential clone.
