SOX9 interacts with a component of the human thyroid hormone receptor‐associated protein complex

摘要: SOX9 transcription factor is involved in chondrocyte differentiation and male sex determination. Heterozygous defects the human gene cause campomelic dysplasia. The mechanisms behind function are not understood despite description of different target genes. This study therefore sets out to identify SOX9-associated proteins unravel how interacts with cellular machinery. We report ability interact TRAP230, a component thyroid hormone receptor-associated protein (TRAP) complex. Both vitro vivo assays have confirmed that detected interaction specific occurs endogenously cells. Using co-transfection experiments, we also shown TRAP230 interacting domain can act dominant-negative manner regarding activity. Our results add list activators communicate general machinery through TRAP complex suggest basis for collaboration coactivators could contact same coactivator/integrator
