摘要: The gonads are responsible for the production of germ cells through both mitosis and meiosis. Skp2 is receptor subunit an SCF-type ubiquitin ligase a major regulator progression into S phase cell cycle, which it promotes by mediating ubiquitin-dependent degradation p27, inhibitor proliferation. However, role Skp2-p27 pathway in development remains elusive. We now show that disruption mice results marked impairment fertility males, with phenotypes resembling Sertoli cell-only syndrome men. Testes Skp2-/- manifested pronounced hypoplasia accompanied massive apoptosis spermatogenic cells. Flow cytometry revealed increased prevalence polyploidy spermatozoa, suggesting aneuploidy these induction apoptosis. Disruption p27 gene restored development, indicating testicular animals attributable to antiproliferative effect accumulation. Our thus suggest compromised cycle caused accumulation gonadal mice. consequent reduction number mature gametes accounts decreased animals. These findings reinforce importance regulation development.
