摘要: The pga1 gene encoding an endopolygalacturonase was isolated from a hyperpectinolytic mutant strain of Penicillium occitanis. It consists ORF 1.155 kbp putative protein 346 amino acids with predicted molecular mass 39 kDa, belonging to the family 28 glycosyl hydrolases. deduced acid sequence comprises 38 N terminal prepropeptide. nature and position comprising active site as well overall three-dimensional structure were conserved between P. occitanis all polygalacturonases. coding region is interrupted by three short introns 57, 53 65 bp in length. In addition determination transcription start site, promoter analysed. showed conservation known response elements for CreA Hap2-3-4 factors. Southern blot analysis at high stringency shows that polygalacturonase exists single copy fungus genome. Northern confirmed constitutive CT1 mutation levels mRNA observed either on pectin or glucose-grown cells.
