Dynamic and stable populations of microtubules in cells.

摘要: Using a new immunocytochemical technique, we have visualized the spatial arrangement of those microtubules in cells that are stable to biotin-tubulin incorporation after microinjection. Cells fixed at various periods time injection were exposed antibody biotinylated tubulin and several layers secondary antibodies; these prevented reaction biotin-containing with antitubulin antibodies. The had not incorporated could then be stained anti-tubulin fluorescent antibody. In BSC1 cells, most cell exchange half-time 10 min. A separate population can detected, using above techniques, for 1 h or more; characteristic pericentrosomal as compared majority dynamic microtubules. Unlike microtubules, nongrowing. average BSC-1 contains approximately 700 microtubules: 500 growing 4 micron min-1, 100 shrinking 20 relatively more exchange. potential significance is discussed.
