Gene structure, molecular characterization and transcriptional expression of two p38 isoforms (MAPK11 and MAPK14) from rock bream (Oplegnathus fasciatus)

摘要: The p38 kinases are one of the four subgroups mitogen-activated protein kinase (MAPK) superfamily which involved in innate immunity. subfamily that includes members namely p38α (MAPK14), p38β (MAPK11), p38γ (MAPK12) and p38δ (MAPK13), regulates activation several transcription factors. In this study, a (OfMAPK11) homolog (OfMAPK14) Oplegnathus fasciatus were identified at genomic level. Results clearly showed both MAPK11 MAPK14 well-conserved structural- amino acid (aa)-levels. Genomic sequences OfMAPK11 (∼ 15.6 kb) OfMAPK14 13.4 had 12 exons. A comparison exon-intron structural arrangement these genes from different vertebrate lineages indicated all exon lengths highly conserved, except their terminal Full-length cDNAs (3957 bp) (2504 encoded corresponding proteins 361 aa 360 aa, respectively. Both OfMAPK harbored Ser/Thr catalytic domain (S_TKc domain) an loop with dual phosphorylation site (TGY motif) specific-binding sites for ATP substrates. Molecular modeling substrate binding rock bream MAPKs revealed conservation crucial residues orientation 3D space. Transcripts OfMAPKs ubiquitously detected eleven tissues examined, however levels. modulation OfMAPKs' upon pathogen-associated molecular patterns (PAMPs: flagellin, lipopolysaccharide poly I:C) pathogens (Edwardsiella tarda, Streptococcus iniae iridovirus) was investigated. Among seven examined tissues, flagellin-challenge upregulated mRNA level head kidney. Meanwhile, expression liver other immune-challenges varied time-dependent manner. Collectively, results suggest true subfamily, might be induced by immune stimuli.
