摘要: To understand how the differentiation of stem cells to oligodendroglial progenitors is regulated, we established cultures neural from neonatal rat striatum in presence epidermal growth factor (EGF) as free-floating neurospheres that were then exposed an increasing amount B104 cell-conditioned medium (B104CM). The resultant proliferated response B104CM but no longer EGF. In vitro analysis and transplantation studies indicated these committed lineage, they thus referred oligospheres. Further characterization their expression early markers, cell cycle, migration, self-renewal suggests pre-O2A progenitors. RT-PCR oligosphere expressed mRNAs platelet-derived alpha receptor addition fibroblast not EGF receptor; latter two by neurosphere cells. Thus, progression likely induced factors B104CM.
