摘要: Layered double hydroxides (LDHs) have been known for many decades as catalyst and ceramic precursors, traps anionic pollutants, catalysts, additives polymers, but they recently attracted attention potential nano-sized carriers therapeutic/bio-active molecules genes. Among the different nanoparticles that shown to facilitate gene and/or drug delivery, LDH are particularly well suited this purpose due their desirable properties. In research Mg2Al(OH)(6)NO3 of varying lateral sizes were synthesized by altering synthesis conditions. The conditions influencing particle size distribution suspensions (a) temperature during co-precipitation step (b) duration hydrothermal treatment association these with plasmid DNA was studied it established that-in contrast previously published reports-for used no significant intercalation occurs. plasmids wrap around individual particles instead aggregation is observed. However, observed strong interaction between DNA, nonetheless evaluated transfection agents mammalian cells. Considerable efficiencies when transfecting adherent cell lines (i.e., HEK293T, NIH 3T3, COS-7, CHO-K1) observed, while suspension CHO-S cells remained unsuccessful. This attributed formation aggregates upon DNA-LDH complex which settle on top constant agitation cultures not surface e.g., (C) 2009 Elsevier B.V. All rights reserved.
