摘要: A rise in extracellular calcium (Ca2+e) suppresses not only secretion of parathyroid hormone (PTH) but also expression the PTH gene to ensure constant plasma Ca2+ level. nuclear protein(s) a wide variety cells bound specific DNA elements (negative responsive elements, nCaREs) human sequence-specific and Ca2+e concentration-dependent manners. Our Southwestern cloning revealed that redox factor protein (ref1), which was known activate several transcription factors via alterations their state, belonged an nCaRE binding protein. The level ref1 mRNA as well its elevated by increase concentration. In gel shift assay, anti-ref1 antibody eliminated formation nCaRE-protein complex. We found there another interacting with nCaREs ref1. Further, experiments antisense-ref cDNA vector introduced into cultured suggested (nCaRE)-ref1 interaction led Ca2+e-mediated transcriptional suppression. Thus, it is concluded possesses repressor activity addition function auxiliary
