摘要: Summary The immunosuppressants rapamycin and FK506 bind to the same intracellular protein, immunophilin FKBP12. FKBP12-FK506 complex interacts with inhibits Ca*+-activated protein phosphatase calcineurin. target of FKBPl2-rapamycin has not yet been identified. We report that a containing 245 kDa 35 components, designated FKBPlP targets 1 2 (RAFT1 RAFTS), FKBPl2 in rapamycin-dependent manner. Sequences (330 amino acids total) tryptic peptides derived from RAFT1 reveal striking homologies yeast TORgene products, which were originally identified by mutations confer resistance yeast. A cDNA was obtained found encode 269 (2549 acids) is 43% 39% identical TOR2 TORl, respectively. propose direct FKBPlS-rapamycin mammalian homolog TOR proteins.
