In vitro pharmacological purging of human bone marrow is enhanced by the use of lonidamine

摘要: Lonidamine (LND), previously reported as a useful antitumor substance in combination with physical or chemical agents, has been studied for its capacity increasing pharmacological elimination vitro of residual tumor cells from human bone marrow. Different drugs were tested association LND against mixtures marrow and cell line, clonogenic leukemic blast progenitors, normal precursors. The results demonstrated that increased the efficacy anthracycline derivatives (Adriamycin, Mitoxantrone) both on line while VP-16 ASTA-Z 7654 was not affected by same substance. toxicity stem reflected each drug modified addition LND. While consistent dose-dependent CFU-GM reduction observed immediately after treatment different drugs, complete recovery reached 7 14 days long-term cultures. Because low certain agents vitro, could play an important role purging prior to autologous transplantation.
