摘要: Often microarray studies require a reference to indirectly compare the samples under observation. References based on pooled RNA from different cell lines have already been described (here referred as RNA-R), but they usually do not exhaustively represent set of genes printed chip, thus requiring many adjustments during analyses. A could also be generated in vitro transcribing collection cDNA clones use T3-R). Here we describe an alternative and simpler PCR-based methodology construct similar (Chip-R), extensively test it both RNA-R T3-R. The Chip-R T3-R dramatically increases number signals slides gives more reproducible results than RNA-R. Each preparation is evaluated simple experiment comparing two populations. Our show that introduction always interferes with analysis. Indeed, direct comparison able identify up- or down-regulated any reference-mediated However, if has used, are guarantee reliable
