Heat shock protein-containing exosomes in mid-trimester amniotic fluids.

摘要: Abstract Exosomes are multivesicular bodies formed by inverse membrane budding into the lumen of an endocytic compartment. Fusion with plasma leads to their release external milieu. The incorporation heat shock proteins exosomes has been associated immune regulatory activity. We have examined whether protein-containing present in mid-trimester amniotic fluid. were isolated from fluids sequential low-speed and high-speed centrifugation followed sucrose density gradient centrifugation. Biochemical characterization included floatation pattern gradients, acetylcholinesterase (AChE) activity Western blot analysis for exosome-containing proteins. each 23 tested. They banded at a 1.17 g/ml positive AChE contained tubulin, inducible 72 kDa protein, Hsp72 constitutively expressed Hsc73; they negative calnexin. Exosome concentrations correlated positively number pregnancies. Heat constituents may contribute regulation within cavity.
