摘要: There is substantial interest in implementing a bioinformatics tool that allows the design of oligonucleotides to support development vitro gene synthesis. Current protocols make long synthetic DNA molecules rely on assembly set short oligonucleotides, either by ligase chain reaction (LCR) or PCR. Ideally, such should represent both strands final molecule. They be adjacent same strand and overlap complementary from second ensure good hybridization during assembly. This implies thermodynamic properties each oligonucleotide have consistent across set. Furthermore, any given has totally specific its target avoid creation incorrectly assembled sequences.
