Factors determining the superior performance of lipid/DNA/protammine nanoparticles over lipoplexes.

摘要: The utility of using a protammine/DNA complex coated with lipid envelope made cationic 1,2-dioleoyl-3-trimethylammonium propane (DOTAP) for transfecting CHO (Chinese hamster ovary cells), HEK293 (human embryonic kidney NIH 3T3 (mouse embryonal and A17 (murine cancer cells) cells was examined. widely used DOTAP/DNA lipoplex employed as reference. In all the tested cell lines lipid/protamine/DNA (LPD) nanoparticles were more efficient in than lipoplexes even though composition same both devices. Physical-chemical properties found to control ability nanocarriers release DNA upon interaction cellular membranes. LPD complexes easily their payload, while remain largely intact accumulate at nucleus. Collectively, these data explain why often exhibit superior performances compared trasfecting represent promising class gene delivery.
