RNAi screening reveals a large signaling network controlling the Golgi apparatus in human cells

摘要: The Golgi apparatus has many important physiological functions, including sorting of secretory cargo and biosynthesis complex glycans. These functions depend on the intricate compartmentalized organization apparatus. To investigate mechanisms that regulate architecture, we developed a quantitative morphological assay using three different compartment markers image analysis, performed kinome- phosphatome-wide RNAi screen in HeLa cells. Depletion 159 signaling genes, nearly 20% genes assayed, induced strong varied perturbations morphology. Using bioinformatics data, large regulatory network could be constructed. Specific subnetworks are involved phosphoinositides regulation, acto-myosin dynamics mitogen activated protein kinase signaling. Most gene depletion also affected particular glycan biosynthesis, suggesting cascades can control glycosylation directly at level. Our results provide genetic overview pathways human
