摘要: Abstract The muscarinic acetylcholine receptor was solubilized from rat brain cortex by zwitterionic detergent 3-[(3-chloramidopropyl) dimethylammonio]-1-propanesulfonate (CHAPS). About 15% of the binding activity and 40% destroyed detergent. Binding antagonist [ 3 H]-N-methyl-4-piperidyl benzilate (4NMPB) saturable. Scatchard analysis revealed a single population sites with K D value 0.7 nM B max 340 fmoles/mg protein. homogenate CHAPS treated pellet soluble receptors showed similar affinity for agonists oxotremorine carbamylcholine antagonists QNB atropine. dissociation 4NMPB appears slightly slower than membrane bound receptors.
